Question I have 1 sys_fan header and 1 cpu_opt, what setup for intake & exhaust fans should I use?

Usually you would want the intake fan to be connected to the CPU-OPT since it most likely has a quicker ramp-up time than the SYS-FAN. You should check your motherboard's bios to see what ramp-up settings are available for both connectors and choose the quickest ramp-up for the intake. And with that motherboard, a higher wattage is available for the CPU-OPT connector, which is another positive factor for the larger fan.
Make and model of your case? Looks like something Thermaltake made about a decade ago. From your picture all I see if a single 200mm fan, which should be 3pin and there are no fans mounted to the rear which I'm assuming is what you're referring to as 80mm mounts. Make and model of your motherboard?

Yes, case is Thermaltake Suppressor F1 (I had an extra one in box until now.) Pic is stock, I have installed an 80mm exhaust fan. This is going to be a gaming pc so I also upgraded the cpu cooler and installed a RTX 4060.
Motherboard is GIGABYTE B760I AORUS PRO LGA 1700 Intel B760 DDR5 SATA 6Gb/s Mini ITX
I see looking at the specs it states has " Smart Fan 6:Features Multiple Temperature Sensors, Hybrid Fan Headers with FAN STOP."
Hopefully this will allow for some better control over the fans in whichever configuration. Thanks!

Both Sys_FAN and CPU_OPT can ramp up immediately either by using FULL SPEED or Manually. I'll put the intake fan on CPU_OPT, right now it's on SYS_FAN and at full speed is 800rpm.
Awesome, so glad I can use both headers for more control over the fans. Thanks for the help, look forward to actually gaming with it and seeing how well the cooling works
