If I format my External HDD to FAT32 to flash my BIOS, will the data on it still be in tact?

I'm borrowing the HDD from a family member, can I just format it back and everything is fine? I've never flashed BIOS before.

Hard Disk Drive: Seagate Barracuda (ST1000DM003) 1TB

I think you misundarstand some basic concepts here.

1st -
Formatting a HDD means a complete erase. Nullify everything. Or in other words - no data survives.

2nd -
A BIOS update does not require an external hard drive. You just need a USB stick usually.
