I'm trying to learn how to setup ISA Server. I have the server setup but
I'm having trouble getting my internal computers to accept email. I created
a protocol filter for port 25 and port 110 but I'm not sure. Can anyone
provide a good source for information. I tried the Microsoft site but I
could find any specific info on settings for internal email clients.

Tom Shinder's ISA Server site is probably the best resource available
anywhere on the 'Net:

www.isaserver.org

Tom has also written a pretty definitive guide to ISA that's published by
Syngress Press - you should take a look at a sample chapter to see if you
want to pick it up for your reference.
