I have a Partition (E) that is used for data files and i have DFS on this
drive. I need to replace these drives with newer ones. My concern is that
how do i go about doing this without affecting users shares. I installed the
new drives and the new and the partition is now drive letter F.

What is the best way to accomplish this?

Use Disk Management to put the new drive back to E:.

The share information is in the registry key HKLM\System\ControlSet00*\services\lanmanserver\shares. You will need to keep the drive letter the same to preserve the paths to the shares
