pci-e lane issue

my motherboard is a S1200BTL with an xeon E3-1220V2. the mobo has 1 pci-e slot x16 and 3 pci-e x4 slots. the x16 is running in x8 mode because of the x4 lanes and the configs my cpu can support. if i disable 2 of the x4 slots, will my x8 slot run in x16 automatically?

No, it will not run at x16, it only runs up to x8 maximum. It is an physical x16 slot wired only as PCIe 2.0 x8.
