Not unless you are trying to use a serial device. They used to be used for mice, old printers, and console devices. They are not commonly used with most modern systems.
Enabling the serial port in your BIOS should allow Windows to detect the device and automatically install drivers for it (assuming this is a port on the motherboard). Is this a add-on card or one built into the motherboard already?
