Ok so I fixed my previous BIOS issue I was having with PC not posting (I had to install BIOS with a clean mobo no CPU, ram or GPU) however I have discovered the 4th slot for ram if I put a stick in there the PC won't post. I have tried several different ram sticks. Will I be ok putting the other ram stick in another slot? I know your mean to have ram in second and 4th slot but if 4th slot broken can I use one of the others available? Or would this cause more problems? and should I just get a bigger single stick of ram?

Thanks in advance.

If 4th is not working I would put sticks in 1st and 3rd. Most motherboards should accept that with no problem, but there are some that might have an objection. No harm trying this though.
Two sticks are usually preferable to one big.

first and third don't work but first and second slot did.. I know its not ideal though.. so maybe third and fourth both broken?

Check the third alone - if it is not working then there is a good chance you have a CPU/socket bent pins (or maybe just a cooler screwed too tight).
yes third one doesnt work on its own 🙁 but first two slots working good and even oc'd them a bit.. WIll i be fine leaving it at this? only a soft oc. The memory is xmp to 4400 but I set to 3400 on higher cl settings.
