RAM Stick Configuration

Is it ok to only use one stick of 16 GB RAM on the GA-Z270P-D3 Mobo?

Yes, just dual channel mode will not be active.

I'm not sure that the GA-Z270P-D3 has a predetermined ideal RAM slot, so realistically, slot 1 or 2 should be fine according to your motherboard manual.
