I'm thinking of getting Samsungs 950 M.2 SSD to use for Windows 10 and a few of my favourite games, but from videos I've seen im a bit confused on how to set it up, I will be using Windows 10 DVD, not usb. Is it possible to use w10 from DVD with the M.2 on a brand new build?

If I recall correctly some mobos also require CSM (Compatibility Support Mode) be turned on.
If you have other drives connected, disconnect them (just easier I find).

I'm also assuming your motherboard ha's a UEFI Bios that allows booting from an NVMe drive?

I had no issues installing windows 10 from a USB drive on to my 950 pro m.2.
It's more a question of how your motherboard recognizes it, and some of your BIOS settings you might need to fiddle with to get it to oot from the m.2 drive first

I'm installing Windows from disc, so after install I'm best of going into bios and selecting the m.2 to be the bootable drive? Sorry this will be my first build in 15 years.
