It won't let you delete it if it's important to for windows to run. Both folders are safe to delete but you might want to keep the backup in case of system re-installing.
It absolutely will let you delete some folders that are important, or more accurately some files that are in use at the time will not be deleted, anything not being used at that time can be deleted (and should not be).
OP you are proposing to delete a backup folder, do you have other backups? is this an old backup? The seagate folder you are planning on deleting probably includes the tools for running backups to an external drive.
I'd therefore leave them alone unless you have other mechanisms in place to create backups.
