I recently upgraded my setup from 16GB of RAM to 32GB and realized that Home Premium can only utilize 16. I have a Windows 7 Installation Disk that has a copy of Ultimate 64Bit in it. Can I upgrade my OS to Ultimate using that Upgrade feature and still keep all my files and programs without issues or should I start searching for installers for software already installed on my PC?

You will need to buy the upgrade. Upgrading from home to ultimate is not free. The upgrade from home to pro is cheaper and pro will be able to use all your new ram.
I've done something similar upgrading from Win7 Professional to Ultimate, and I did it by entering my Ultimate product key into Windows Anytime Upgrade; it accepted the key no problem, downloaded some stuff, and a few hours later I was on Ultimate edition and all of my installed programs and files were intact. Do you have a product key for Ultimate? If so, you don't even need the installation disk.

I don't have a key for Ultimate, just the USB stick I got from a friend for the copy of Windows.
