It is ok to remove And reposition the cpu cooler on a freshly apply termal paste?

You may introduce microscopic air bubbles into the paste, if you do. That could inhibit cooling performance. But use something* to smear it around both surfaces before you are ready to re-assemble the cooler, and you should be OK.
*Your finger in a clean plastic baggie will work.
