I have the Asus Prime Z270-P motherboard and I am thinking about getting a new m.2 nvme ssd for my pc. However I do not know if when I put an nvme drive in, if it will slow down my GPU in my top 16x slot. Thank you for any replies in advance.

No. Your GPU will continue to have 16 PCIe lanes. Even if your PCIe lanes were cut in half you wouldn't notice the difference.
