In theory, no. As long as it is of good quality; one that doesn't add impedance due to poor contacts or wiring. Here's someone that says he's done testing with risers/extenders: http://www.overclock.net/t/1427731/pci-express-extender-cables-benchmarked
These were tested a few years ago with a somewhat older system, but I imagine the results with modern cards and systems would still be the same.

You lose a little bit. I helped a buddy do risers on his wall mounted system. Some work better than others, but even with the really good risers, there was about a 5% - 10% loss on Firestrike benchmark frame rates.

Notice that the author says he even daisy-chained the 300mm riser into the 200mm riser, and he still got nearly identical scores. Even an insignificantly bit higher in Firestrike than with direct to the slot.
