For a great step by step in how to assemble your PC get hold of this PDF from
MSI [motherboard and component builder].

http://www.msi.com.tw/html/support/manual/note/quick_installation.pdf

You will need to look at your ASUS manual and look to the BIOS setup for any
changes needed with the change of CPU. Some have fully automatic
configuration, others need things such as CPU Clock, FSB etc.

As for thermal contact and cooling: a new CPU should come packaged with the
correct cooling device. It has the thermal contact material already affixed
to the CPU and so all it takes is to unpack: insert on MOBO: connection of
Fan. Easy as 1-2-3.

The CPU only fits one way, look at the pins and you'll see
that one corner is different than the others. Also on top
there should be a dot on that corner. Lift the lever on the
socket and the CPU should drop in by its own weight. put
the lever down and locked after the CPU is fully down.
You need a thermal paste to make heat transfer, just a small
drop spread about 2/1000" thick with a clean credit card
edge.
