How to convert toshiba external drive file format from fat32 to ntfs

Hello,
i have a toshiba external hard drive of 500GB. i wish to back up my computer but the PC on vista has an NTSF platform while the ext HDD FAT32. how do i convert the ext hdd to NTSF without losing the stored data? will the conversion affect it's functionality?

I have used the convert command once going from FAT to NTFS and it worked flawlessly and no data was lost. I really can't see the need for a 3rd party application.
