I had run android iso "lollipop_5.1_3.18.8_x86_20150417" on my usb drive which is of 16gb but I cannot preserve or save files across reboots. Please help!
How do you run that "iso" file?
If you boot off it (as you would for Live Linux), ISO is read-only medium (even if you read it off USB), so you cannot save anything on it. Look for way to actually "install" that ISO onto the USB.

Is this your "Android ISO"? There is a special section called "USB Install"
You need two USB sticks: One temporary (where you put the ISO file and boot from), and one where to actually install that Android, and from which you would boot later on.
I would highly recommend disconnecting all of your hard drivers while doing that!!
