Question PC won't boot ssd.

I recently upgraded my motherboard to an Asus TUF B450-m Plus Gaming, I install the motherboard and the rest of my parts and the first boot was fine. I turned it off and close the case up and since then have not been able to boot through my main drive (Samsung 970 m.2 nvme 500gb ssd) this drive already has windows 10 and all my other crap on it is not recognised in the bios.
I tried countless different things but cannot get it to boot through my ssd.

Motherboard: Asus TUF B450-M Plus Gaming
CPU: Ryzen 5 2600 at base speeds.
GPU: Inno3D GTX 1060 6 GB
Ram: 8GB of crappy geil ram 2400mhz
SSD: Samsung 970 Evo m.2 nvme 500gb
HDD: 2TB Toshiba, it's old but works fine.
PSU: Corsair VS650

unplug the hdd? does bios see the ssd?
Im in the bios with only my ssd in the system. In the boot priority section I have windows boot manager (Samsung ssd etc) but in my data information all ports includinh m.2 are N/A. So it notices my ssd for boot but doesn't think it's in my pc.????
