Question PCIe stopped working, is it my mobo?

Hi guys, yesterday i was using my pc and everything was working fine as always, but today i had a problem, here are my specs:

Asus tuf b450 plus gaming
GTX970
Ryzen 2600
CORSAIR Vengeance LPX 16GB DDR4

So today when i tried to turn my pc i heard the POST single beep but nothing was showing up on the screen, so after i took the case off and checked everything was ok i tried my GPU on another pc and it worked, after that i put the GPU on the PCIe slot 2 and everything worked fine again. So, do you think that my mobo is faulty? Is there any other tests i can do?

Check BIOS and see if PCIe_1 is default.
