How do you test if a PCIe slot is x16 lanes?

You can start by looking at it. It will be physically as large as a graphics card PCIe edge. BUT that only is x16 form factor. To determine the number of active lanes, you have to look at the motherboard specs. You may have to look at the motherboard in conjunction with the CPU choice since SOME Intel CPUs have more PCIe lanes than others.
