Will an AMD R9 380 4GB have an advantage over the 2GB version in harder to run games such as GTA V?

Right now it doesn't really need that memory, its used for textures. However games will be released that can use more and more memory soon, so its only a matter of time before there CAN be an advantage to the larger memory. For the small price difference its worth it on that fact.

It takes some of the newest games at 1440p with HD texture packs to use more than 2GB of vRAM.

You will probably be fine.

But if the price difference isn't too much the 4GB card would be a good idea.
