Will 1600MHz DDR3 RAM work on Asus M5A78L-M/USB3 motherboard and AMD FX 6100 without overclocking?

Your mobo spec says " AMD FX Series CPU on this motherboard supports up to DDR3 1866MHz as its standard memory frequency." so YES.

But also says (O.C.) overclocking, I guess. Is it have something to do with?

Probably means that the ram will start at a speed of 1333mhz in the bios and needs manually be set to the rated speed and timings.
