When the MCT is showing Windows 11, I plan on using it to create the Windows USB 11 media.
Do I need to perform an update first or can I just boot from the usb for a complete installation and activation?
I have the 25 character key if necessary?

IF its anything like win 7 to 10 upgrade, you need to update to win 11 first, and then once license is converted, can clean install

You shouldn't need the key once its activated as Windows 11.

We don't know if ISO will be available right away as MS must know most tech savvy people will use that way.

Supposedly, the only official path is an Upgrade to be available on Oct 5.

Fresh install not until early next year.

My understanding from reading indicates that the version that is out now to the "preview" people is likely the same as the official release will be. Indicated that some of the other sections of the OS have come online, and that any computer that meets requirements will update to the release revision as soon as possible after official release date. This would indicate that even the commonly available .iso that is floating around ATM will become the release edition as quickly after that date as the (surely) overburdened servers can get to you. The same article mentioned that it's highly probable that many users who have the correct hardware and will be eligible may well have to wait some time for the "upgrade to" to appear within the Windows Update console simply due to demand.

Exactly what happened with Win 10.

Not every one and every system got the "Update Now" thing at the same time.

Two other forums state you do not have to upgrade just make the MCT and clean install.
Is that with the current Insider Preview, or on actual release day?

I prefer info direct from the source:

"Today, we are thrilled to announce Windows 11 will start to become available on October 5, 2021. On this day, the free upgrade to Windows 11 will begin rolling out to eligible Windows 10 PCs and PCs that come pre-loaded with Windows 11 will start to become available for purchase. "

At my level I cannot fully utilize the firewall and other features. TPM and secure boot keep giving me trouble.
I need a suite that will do things for me. I always take you advice so Defender it is.
