Question Main PCIe x16 slot is not working on startup on ASUS X570 ?

I am looking for any advice on how to resolve or further test this issue. I have a digital multimeter, but don’t know how to use it on a mobo. I am also willing to try software/bios solutions. Any suggestions are much appreciated.

My GPU doesn’t work when plugged into the top pcie x16 slot on my asus x570 mobo - nothing comes on screen when starting the pc and I get the 1 long beep and 3 short beeps. I have tested a backup GPU and get the same result. Either GPU works fine in the second pcie x16 slot. This has been the case since I first got this mobo, and I just ran my GPU in the second slot and didn’t worry about it. Now, I have a new case and using that bottom slot is rough for airflow purposes. AND I just learned that the top pcie slot is superior to use over the bottom one. So, now I am more determined to figure out why the top slot is not working.
With both GPUs installed at once, the system starts up fine and just only recognizes the GPU in the bottom slot. Though, the GPU in the top slot will light up its own rgb.

I have read some posts and tried the following: updating my bios, disabling CMS, and forcing that slot to use gen 3 instead of auto. Nothing has worked so far for either GPU to get past the post.
I tried calling ASUS and they said they cannot provide any troubleshooting over the phone. They said my only option to get help was to ship the mobo to them for testing in their facility, and there would be shipping / repair charges.

Mobo: Asus Tuf gaming x570-plus wifi
CPU: AMD Ryzen 7 3700X
GPU: Gigabyte Eagle - Nvidia Geforce RTX 3070Ti
Secondary GPU for testing: MSI - Nvidia N740
Ram: G.Skill 3600 32GB 8x4.

Should have claimed warranty first time you found the problem. PCIe slot can be faulty just like any other part. Multimeter is not going to help any.
 
Yes, in hindsite I could have returned it. When I have 2 GPUs inserted, the system will use the GPU in the second (bottom) slot. I can see the top slot GPUin the device manager an it just says this cannot be used due to resource restrictions (error code 31 if I remember correctly). If the PCIE slot was bad, would this GPU come up in the device manager at all?

PCIe x16 has 82 pins and bunch of circuits serving them, any one can cause problems.
